RESUMEN

The objective of this study was to determine the clinical applicability of maternal intrapelvic area (PA) and selected morphometric parameters that can be measured before parturition in predicting dystocia in dairy heifers. The measurements were performed in 374 late-gestation Holstein-Friesian heifers. Inner pelvic height and width were measured using a pelvimeter, and PA was calculated. The heifers were monitored continuously around the time of calving, and calving difficulty was categorized as: unassisted calving (UC), slight assistance (SA), considerable difficulty (CD) and veterinary assistance (VA). Calving performance was analysed with the χ2 test, and the effect of body dimensions on the course of parturition was evaluated by one-way analysis of variance. Dystocia (CD + VA) was predicted with the use of the classification tree method. Dystocia accounted for 29.14% of all deliveries. The percentages of stillbirths and retained placenta increased (p < .01) with increasing calving difficulty. Average PA immediately before parturition was smaller (p < .01) in group VA (223.2 cm2 ) than in group UC (253.3 cm2 ). According to the classification tree, dystocia may occur (74.07% odds) in heifers with PA < 254.2 cm2 and a rump angle <5.68° before parturition. Measurements of heifer's cannon circumference and sire's body size improve the accuracy of dystocia prediction.

This was a prospective cohort study to determine how events from birth until first calving affect reproductive performance in the first lactation in pasture-based dairy herds in Victoria, Australia. Events during the preweaning (0 to 84 days), weaning to first breeding (85 to 473 days) and first breeding to first calving periods (474 to 804 days) were recorded and their association with reproductive performance during the first lactation was quantified. Reproductive performance outcomes included the number of days from first mating start date to first service (MSD-S1) and the number of days from first mating start date to first conception (MSD-CON). Predictors for reproductive performance included: passive transfer status as a calf; season of birth; age and breed of the dam; breed; the presence or absence of dystocia at the time of the heifer's birth; active feeding of colostrum versus being left on the dam for colostrum intake; presence of twinning; the presence or absence of preweaning diarrhoea; preweaning, prepubertal and postpubertal average daily gain; the presence or absence of periparturient disease at first calving; age at first calving; body condition score at first calving; sex of the first-born calf; the presence or absence of a stillborn calf at the first calving and requirement of assistance at first calving. Two Cox proportional hazards regression models were developed: the first for early life event variables associated with MSD-S1 and the second for early life events associated with MSD-CON. Heifers born in autumn and winter had 2.89 (95% CI 1.50 to 5.59, P = 0.002) times and 1.97 (95% CI 1.12 to 3.44, P = 0.018) times the daily hazard of being inseminated compared with heifers born in spring, respectively. For the MSD-S1 analyses heifers that gave birth to a live calf had 1.64 (95% CI 1.14 to 2.36, P = 0.008) times the daily hazard of being inseminated compared with heifers that had a stillborn calf. Increases in weight gain during the first breeding to first calving period by 0.1 kg/day increments increased the daily hazard of first insemination by a factor of 1.10 (95% CI 1.00 to 1.22, P = 0.043). Heifers that experienced periparturient disease had a significantly lower hazard of conception per day compared with heifers that did not experience periparturient disease at the first calving (HR 0.67, 95% CI 0.50 to 0.91, P = 0.009). Increases in weight gain during the first breeding to first calving period by 0.1 kg/day increased the daily hazard of conception by a factor of 1.10 (95% CI 1.01 to 1.21, P = 0.038). We conclude that of all the growth periods assessed in this study, events that occurred during the first breeding to first calving period (474 to 804 days) had the greatest association with reproductive performance in the first lactation. There should be a focus on increasing growth rates during this period and reducing the risk of stillbirth and periparturient disease to improve reproductive performance in the subsequent mating period after calving.

The Black-and-White (BW) breed, which until recently had dominated in Europe, was replaced by the Holstein-Friesian (HF) breed. As a result, the incidence of dystocia has increased. Dystocia occurs most frequently in heifers, and it is associated with high calf weight and/or too narrow pelvic openings in heifers. The aim of this study was to evaluate retrospectively the effects of pelvic dimensions and rump angle on calving ease in two cattle breeds. The research was carried out in four barns where BW and HF cattle were used. The course of parturition was evaluated in 317 heifers (BW, n=169; HF, n=148) based on direct observations. Calves were weighed, external and internal pelvic measurements were performed (using the Rice pelvimeter), and rump angle was determined in heifers. Based on the course of parturition, heifers of both breeds were divided into easy calving (EC) and difficult calving (DC) groups. The frequency of DC was 24.3% in HF heifers and 13.1% in BW heifers. In comparison with DC heifers, EC heifers had a larger pelvic area, in particular the internal dimensions of the bony pelvis, and a higher rump angle. In comparison with BW heifers, HF heifers had a smaller rump angle, a narrower pelvis and a lower ratio of pelvic area to calf weight. High dystocia rates in HF heifers could result from a relatively large fetus size and a less preferable pelvic size and rump angle. High variation in the internal pelvic dimensions in HF heifers indicates that the incidence of dystocia can be reduced through selection for a larger pelvic size and the optimal rump angle.

A retrospective study was conducted to evaluate temporal patterns of environmental heat stress during the in-uterus period of development (IUP) and the 3-month post-natal (PN) period of dairy heifers, and to estimate their association with the age at first calving (AFC). Data from 30 dairy herds in Northern Greece including 9098 heifers were extracted from National Cattle Database. Data (2005-2019) regarding 230,100 farm-specific ambient daily temperature and relative humidity records, were obtained from ERA5-Land. Average monthly Temperature-Humidity-Index values (THI; low≤68, and high>68) were calculated and matched for each heifer to their IUP and PN. Subsequently, Cluster Analysis was used with monthly THIs as predictors to allocate heifers to THI clusters. The association of clusters with AFC was assessed with Generalized Linear Mixed Model analysis, an extended form of multiple linear regression. Finally, 8 Heat Stress Clusters (HSC; namely HSC-1 to HSC-8) were identified. Compared to HSC-8 (8th-9th IUP months and 1st PN month) heifers of HSC-5 (4th-7th IUP months) and HSC-6 (6th-8th IUP months) calved 13.8 and 17.8 days later, respectively (P < 0.01-0.001). Moreover, when AFC was treated as a binary variable, heifers of HSC-5 and HSC-6 had 1.15 and 1.34 (P < 0.01-0.001) higher risk of calving for the first time later than 787 days compared to HSC-8, respectively.

Dairy cattle face a variety of stressful events on a daily basis. More specifically, climate change has resulted in more frequent heat stress events that increase the incidence of chronic bacterial infections by inducing conditions like leaky gut syndrome, whereby the integrity of the intestinal epithelium is compromised allowing for luminal bacterial lipopolysaccharide (LPS) endotoxin to infiltrate the host's bloodstream resulting in acute or chronic systemic stimulation of the innate immune system. Repeated exposure to LPS over a short period of time is reported to induce immunotolerance within the host. This LPS tolerance is an essential immunohomeostatic response that can protect against over activation of the inflammatory response during subsequent exposure to LPS. In the present study, Holstein calves (n = 20) were initially stress challenged with either saline, or 100, 200 or 400 ng/kg of LPS administered intramuscular, and again re-challenged with 200 ng/kg of LPS 2-weeks later. Serum was collected every 2 hr for 6 hr to profile changes in circulatory stress biomarkers after the repeated LPS exposures. Heifers that were initially challenged with 100, 200 and 400 ng/kg of LPS demonstrated significantly attenuated cortisol responses in the second challenge (p < 0.01, 0.01 and 0.05, respectively), whereas control animals who previously received saline demonstrated a strong cortisol response at 2 hr after receiving 200 ng/kg of LPS (p < 0.05). The cytokine/chemokine (IL-6, CCL2, CCL3 and CCL4) responses were also attenuated during the LPS rechallenge (p < 0.05). Finally, microRNA expression profiles were determined to assess the epigenetic response to repeated LPS exposure. Interestingly, miR-31 and miR-223 were downregulated in response to the second LPS challenge. The present study demonstrates the dynamic nature of the stress response in dairy cattle as it relates to the development of LPS tolerance. Understanding the roles of various stress biomarkers in the context of innate immune cell tolerance is essential for evaluating their impact on immune system homeostasis.

The incidence of paramphistomosis, caused by the rumen fluke, Calicophoron daubneyi, has greatly increased within Europe in the last 15-20 years. However, the production impacts of this disease are poorly understood. This study firstly aimed to investigate the prevalence of rumen fluke in England and Northern Ireland (NI) by conducting an abattoir survey of dairy and beef cattle which also allowed the impact of rumen fluke on carcass weight, conformation and fat classification to be assessed. Secondly, an experiment aimed to assess the impact of C. daubneyi infection on diarrhea score, production loss and welfare in dairy heifers, while also evaluating the impacts of treating infected heifers with oxyclozanide. Rumen fluke prevalence was greater in NI than in England, with 53.8% (95% CI 51.9 - 55.9%) of the NI cattle carcases sampled being infected compared to 16.3% (95% CI 15.8 - 16.8%) and 17.9% (95% CI 17.4 - 18.4%) detected at the two abattoirs in England. However, there was no significant difference (P > 0.05) in the cold carcass weight between infected and non-infected cattle. Similarly, carcass conformation and fat classification were unaffected (P > 0.05) by the presence of rumen fluke. In the second experiment, daily live weight gain (DLWG), diarrhea score and welfare score were also unaffected (P > 0.05) by rumen fluke infection and by oxyclozanide treatment against rumen fluke. The farms in this experiment were managed to a high standard and animals had no intercurrent disease. Therefore, these findings suggest that on well-managed farms, production losses (growth rates) should not be compromised as a result of sub-clinical rumen fluke infection.

Repeat breeding is a substantial problem in heifer and cow breeding leading to greater infertility for female dairy herds. The aim of present study was to investigate the impact of corpus luteum (CL) presence and category and the first gonadotropin-releasing hormone (GnRH) administration concurrent with exogenous progesterone (P4) treatment on the largest follicle (LF) size and pregnancy rate (PR) in repeat-breeder crossbred dairy heifers submitted to the fixed-time artificial insemination (AI) protocol. Heifers (n= 243) were synchronised with (+GnRH) or without (-GnRH) first GnRH in the 7-day P4-GnRH-prostaglandin F2α-based programme. Each GnRH group was divided on presence of CL into two groups (+CL and -CL) in a 2 × 2 factorial arrangement. The PR was similar among -GnRH-CL (20.7%), -GnRH+CL (68.8%), +GnRH-CL (30.4%), and +GnRH+CL (68.3%) groups. However, presence of CL in heifers produced a 43.6% increase in PR compared to PR of heifers without CL (odds ratio = 6.550). Heifers bearing large-sized CL had greater large-sized LF on the day of fixed-time AI and PR. Plasma P4 concentration was positively related with CL diameter (r= 0.845; p < 0.001). The diameter of ovarian LF on the day of fixed-time AI was positively associated with P4 concentrations (r= 0.512; p < 0.001). We highlight that ovarian CL presence and category at the time of exogenous P4 treatment alters pre-ovulatory follicle size and PR but not initial GnRH treatment in repeat-breeder crossbred dairy heifers submitted to service with the 7-day fixed-time AI programme.

Dairy cattle routinely face a variety of stressors. For example, climate change has resulted in more frequent heat stress events that increase the incidence of bacterial infections by inducing conditions like leaky gut syndrome, whereby the integrity of the intestinal epithelium is compromised allowing for luminal bacteria and their membrane components to infiltrate the host's bloodstream resulting in systemic inflammation. Lipopolysaccharide (LPS) is a well-characterized and biologically relevant microbe-associated molecular pattern (MAMP) that makes up the outer membrane of pathogenic and commensal Gram-negative bacteria and is known to contribute to inflammatory disorders including mastitis, acidosis and septicemia. In the present study, Holstein heifers (n = 20) were randomly allocated into different treatment groups receiving saline, 100, 200 or 400 ng/kg of LPS intramuscularly to create an experimentally induced endotoxemic state. Serum was collected hourly for 8 hr and then again at 24 hr to profile changes in circulatory stress biomarkers. All LPS -challenged animals demonstrated distinct cortisol responses 2 hr post-LPS challenge, and in the 200 ng/kg and 400 ng/kg of LPS treatments cortisol concentrations remained significantly induced for up to 4 hr. Rectal temperature was significantly increased for heifers challenged with 100 and 200 ng/kg of LPS at 2 and 4 hr as compared to their pre-challenge temperature. All LPS-challenged animals demonstrated marked leukopenia and thrombocytopenia as compared to the saline control animals. A total of 8 cytokines, including TNFα, and IL-10, were found to be induced between 2 and 4 hr. Finally, we report that miR-1246, miR-223, miR-29 and miR-31 were significantly induced in animals challenged with LPS as compared to the saline controls. The present study demonstrated that the stress response in dairy heifers is dynamic and there are peak windows of time when cortisol, cytokines and also miRNA are induced, and blood cells are sequestered as part of the systemic inflammatory response. Variability in the response to LPS warrants further investigation in dairy cattle to better understand the contribution of genetics and associations between LPS-induced stress and health and performance.

The present study aimed to compare the growth rate, feeding behavior, reproductive development, and concentrations of GH and leptin in Sahiwal heifers exposed to direct bull contact, bull contact through a fenceline, or isolated from bulls. Overall, 24 heifers were allotted to three treatment groups (n = eight/group) based on birth weight, body weight, and age. Heifers from the non-exposed group (NBE) remained isolated from bulls; heifers stimulated by fenceline bull contact (FBE) remained continuously exposed to bulls through a fenceline, and heifers with direct bull contact (DBE+FBE) had direct contact with a bull during 6 h/day plus continuous fenceline bull exposure for the entire 24 h period. It was considered that heifers attained puberty when progesterone concentration was >1 ng/mL. Biostimulated heifers achieved puberty at both a lower age and body weight (P < 0.05) than NBE heifers. Overall, average daily gain (g/day) was greater in DBE+FBE (516.3 ± 4.5) and FBE (501.6 ± 4.3) than in NBE (441.8 ± 2.9, respectively; P < 0.01). Leptin and growth hormone concentrations were significantly greater in FBE and DBE+FBE than NBE heifers. Both groups of biostimulated heifers devoted more time eating than NBE heifers, P < 0.05). In conclusion, biostimulation of Sahiwal heifers, either by exposing them to bulls through a fenceline or by both fenceline exposure and direct contact, reduced the age of puberty and increased growth performance. Overall, fenceline bull exposure can be recommended as an easy handling management to stimulate reproductive precocity and growth rate in heifers.

In this study, we aimed to comprehensively evaluate the effects of prepartum replacement of corn silage and distillers' grain diets with hay on the postpartum health and production performance in dairy heifers. Pregnant Holstein dairy heifers were randomly assigned to two groups and fed different diets with the same net energy and protein concentrations. The low corn silage plus hay (CH) group diet mainly contained oat hay (28.5%), corn silage (25%) and folium ginkgo (23%). The high corn silage plus distillers' grains (CD) group diet contained corn silage (53.5%), distillers' grains (5%) and folium ginkgo (18%). Dairy heifers were fed from the beginning of pregnancy to the end of the transition period. Calf birthweight, postpartum morbidity, milk production and reproductive performance were determined. Compared with the cows in the CH group, those in the CD group presented increased calf birthweight and a high risk of some postpartum disorders (p < 0.05), particularly ketosis (37.63% vs. 24.54%) and retained placenta (9.41% vs. 3.92%). However, there was no difference between the groups in reproductive performance, including first timed artificial insemination (TAI) rate, date of TAI and conception rate of TAI. The lactation curves showed no difference in milk yield during the peak lactation period of primiparous cows between the CH and CD groups; the total milk production was 33.39 kg and 33.19 kg respectively. Notably, the time to peak milk production was delayed in the CD group. It can be concluded that replacing corn silage and distillers' grains with hay can reduce postpartum morbidity in dairy heifers and shorten the time to peak milk production.

The aim of this study was to compare the health status of contract- vs. conventionally reared replacement dairy heifers over a 2-year period. A total of 120 dairy farmers were enrolled in the study in spring 2018: 55 farmers were rearing their own heifers (control farmers; CFs), and 65 were sending heifers to a contract-rearing farm (source dairy farmers; SDFs). Between spring 2018 and autumn 2019, approximately 5500 replacement heifers from these farms were monitored for signs of ill health during four farm visits using a modified version of the Wisconsin calf health scoring system. Additionally, faecal and nasal swabs were taken from a proportion of heifers with clinical signs of diarrhoea and respiratory disease to determine the associated aetiological agents. Results indicate few differences in the health status and pathogen exposure status of home-reared versus contract-reared heifers. Additionally, the number of source dairy farms represented and commingling of heifers from multiple origins at the rearing unit were not associated with an increased incidence of respiratory disease or diarrhoea among contract-reared heifers. It was concluded that contract-rearing did not result in adverse health outcomes for replacement dairy heifers. This is the first study to demonstrate this finding in a robust, longitudinal, herd-level population study.

Several models for predicting dry matter intake (DMI) of replacement dairy heifers have been developed; however, only a few have been evaluated using data from heifers of different breeds raised under tropical conditions. Thus, the objective of this study was to evaluate the DMI equations for dairy heifers managed under tropical conditions. A total of 230 treatment means from 61 studies using dairy heifers (n = 1513 heifers, average body weight = 246 kg) were used. The animals were grouped into two groups based on their genetics: (1) Bos taurus (Holstein, Jersey, Brown Swiss, and Holstein × Jersey) and (2) crossbred (Bos taurus × Bos indicus). Seven previously published DMI equations (HH, HHJ, QUI, STA, 2001 NRC, OFLin, and OFNLin) for heifers were evaluated using mean bias, slope bias, mean squared prediction errors (MSPE) and its decomposition, and other model evaluation statistics. For Bos taurus heifers, our results indicated that OFNLin and HHJ had lower mean bias (0.13 and 0.16 kg/d, respectively) than other models. There was no significant slope or mean bias for HHJ and OFNLin (p > 0.05), indicating agreement between the observed and predicted DMI values. All other models had a significant mean bias (p < 0.05), whereas the QUI model also presented a significant slope bias (p < 0.02). For crossbred heifers, the STA equation was the only one that did not present mean and slope bias significance (p > 0.05). All other DMI models had significant mean bias when evaluated using crossbred data (p < 0.04), and QUI, OFLin, and OFNLin also presented significant slope bias (p < 0.01). Based on our results, predictions from OFNLin and HHJ best represented the observed DMI of Bos taurus heifers (MSPE ≤ 1.25 kg2/d2, mean bias ≤ 0.16 kg/d), whereas STA was the best model for crossbred heifers (MSPE = 1.25 kg2/d2, mean bias = 0.09 kg/d). These findings indicate that not all available models are adequate for estimating the DMI of dairy heifers managed under a tropical climate, with HHJ and OFNLin for Bos taurus and STA for crossbreds being the most suitable models for DMI prediction. There is evidence that models from Bos taurus heifers could be used to estimate the DMI of heifers under tropical conditions. For heifer ration formulation is necessary to consider that DMI is influenced by breed, diet, management, and climate. Future work should also include animal genetic and environmental variables for the prediction of DMI in dairy heifers.

The objective of this study was to compare the economic performance of an ear tag automated activity monitor system (AAM) versus a timed-AI (TAI) protocol in Holstein heifers. In total, 340 heifers were enrolled onto the study at 13.5 mo of age and randomly assigned to receive either an AAM (n = 170) or TAI (n = 170) protocol before breeding eligibility (D 0). Heifers in the AAM group were fitted with an ear tag AAM and bred based on high activity alert from the system. Heifers in the TAI group received a progesterone releasing intravaginal device on D -8, followed by device removal and prostaglandin on D -3 and gonadotropin-releasing hormone with TAI on D 0. In both treatments, the majority of heifers received sex-sorted semen for the first AI and conventional semen for subsequent AIs, with three opportunities to become pregnant. All heifers were diagnosed for pregnancy approximately 25 d post AI using transrectal ultrasonography, with confirmation at 30 and 45 d. Non-pregnant heifers in the TAI group, were resynchronized using the same TAI protocol. A partial budget was used to compare the costs and benefits of switching from a TAI to an AAM protocol in heifers, including protocol, labour, and rearing costs for each treatment, as well as estimated calf and milk value. Sensitivity analyses were also conducted to determine the effect of pregnancy per AI (P/AI), outsourcing AI, AAM tag cost and herd size on the net outcome. There was no difference in overall P/AI or days to pregnancy between treatments. However, number of AI was greater in the TAI than the AAM group. For the first AI, the P/AI was less in the TAI compared to the AAM group; however, the interval to first AI was less in TAI. There was minimal difference in performance for the second and third AI. There was a net gain of $11.97 per heifer when switching from a TAI to AAM protocol, due to the increased P/AI to the first AI and reduced cost of hormones. Several variables in the sensitivity analyses affected the net outcome. Considering only the first AI, switching to an AAM collar and a larger herd size all increased the net gain. Considering a greater P/AI to the first AI in the TAI group, outsourcing AI, using more AAM ear tags, and smaller herd sizes resulted in a net loss when switching from TAI to AAM. The AAM system resulted in exceptional P/AI and may be an economically viable alternative to improve heifer reproductive efficiency in herds with suboptimal P/AI to TAI.

Our aim was to evaluate the effects of progesterone supplementation after fixed-time artificial insemination (FTAI) on physiological and reproductive variables and serum thyroid hormone concentrations of cooled Holstein heifers during a hot summer season. Sixty-nine Holstein heifers were randomly assigned to three treatments: (1) heifers under visual estrus detection and inseminated according to AM-PM rule (n = 23; control (C)), (2) heifers subjected to FTAI after estrus synchronization using a CIDR insert (n = 24; FTAI group), and (3) heifers treated as the FTAI group plus progesterone supplementation between the day 4 and 14 post-insemination with a reused CIDR device (n = 22; FTAI+SP4 group). All heifers were cooled daily with misting and fans. Physiological variables were measured; likewise, blood samples were collected to determine serum progesterone, thyroxine, and triiodothyronine concentrations. Both respiration rate and rectal temperature were similar among treatments. Conception rate was greater (P < 0.05) in C (65.2%) and FTAI+SP4 (59.1%) heifers than in FTAI heifers (33.3%). Serum triiodothyronine and progesterone concentrations of FTAI+SP4 heifers were similar to those of C heifers but greater (P < 0.05) than those of FTAI heifers. In conclusion, progesterone supplementation post-insemination increased conception rate and serum progesterone concentrations without affecting thermoregulation capacity in Holstein heifers subjected to FTAI and cooling during a hot summer season.

We studied the effects of a hydrophobized reversed-phase feed adsorbent in the form of a polyoctylated polysilicate hydrogel (POPSH) on productivity indicators, metabolic adaptation, and on the level of polychlorinated biphenyls (PCBs) in the blood of growing Holsteinized black-and-white heifers during the transition period. Two groups of two-month-old heifers of 20 head each were used. The experimental group received POPSH in addition to the main diet. The use of the adsorbent led to an increase in daily weight gain by 19.9% and to a decrease in the total concentration of PCB congeners found in whole blood by 40%. The greatest decrease in concentration (35-52%) was observed for tetra-, penta- and hexachlorobiphenyls. These results demonstrate the possibilities of effective protection of calves from lipophilic toxins of feed and their active decontamination.

The present study was aimed at evaluating the differences of ovarian follicular dynamics and circulating progesterone (P4) concentrations between crossbred Holstein heifers that ovulated and did not ovulate after a P4-based synchronization protocol. Twenty-one crossbred (Holstein × Thai native) heifers with random stages of the oestrous cycle were subjected to the ovulation synchronization protocol, using an intravaginal P4-releasing device (Eazi-Breed CIDR®) for 7 days. Out of 21 CIDR-treated heifers, 14 ovulating heifers were classified as the ovulatory group and 7 non-ovulating heifers were considered the anovulatory group. The heifers having new wave emergence in ovulatory and anovulatory groups were 11/14 (78.6%) and 4/7 (57.1%), respectively. In ovulating heifers, the mean (± SEM) diameter of preovulatory follicle (PF, mm) was significantly larger, compared to non-ovulating heifers (7.21 ± 0.32 versus 4.04 ± 0.44; P = 0.001), while the mean (± SEM) follicular growth rates (mm/d) in non-ovulating heifers tended to be lower, compared to ovulating heifers (0.73 ± 0.17 versus 1.06 ± 0.08; P = 0.07). The mean (± SEM) circulating P4 concentration (ng/ml) throughout the CIDR protocol (0-10 days) in non-ovulating heifers was significantly higher, in comparison with ovulating heifers (2.82 ± 0.27 versus 1.83 ± 0.16; P = 0.02). However, no significant difference in the mean corpus luteum volume between groups was observed. In conclusion, the present results suggested that elevated circulating P4 concentrations and smaller PF diameters could cause ovulation failure in crossbred Holstein heifers, following a 7-day CIDR-based synchronization protocol.

The aim of the current study was to investigate the effects of the prepartum external treatment of teats with a combination of four lactic acid bacteria strains viz. Lactobacillus (Lb.) rhamnosus ATCC 7469, Lactococcus lactis subsp. lactis ATCC 11454, Lb. paracasei 78/37 (DSM 26911), and Lb. plantarum 118/37 (DSM 26912) on the postcalving udder health of dairy heifers. The study used a split-udder design. Two weeks before the expected calving date, one of two contralateral teats of a teat pair was dipped with an aqueous suspension of lactic acid bacteria (final bacterial counts 8.40-8.47 log10-transformed CFU/mL) once in a week until calving; the other teat of the pair was not treated. After calving, quarter foremilk samples were taken and investigated cyto-microbiologically. In total, 629 teat pairs of 319 heifers were included. There was an association between the treatment and intramammary infections caused by the major udder-pathogenic bacteria Staphylococcus aureus, Streptococcus dysgalactiae, and enterococci, as well as clinical mastitis in the first 100 days after calving. The present study indicates that intramammary infections with major pathogens and clinical mastitis may be prevented by regular prepartum external application of lactic acid bacteria in dairy heifers.

This study examined the long-term effects of failure of passive transfer of immunity (FPT; diagnosed at 1-8 days of age) on subsequent milk production, growth, reproduction, and lactation performance in dairy heifers from 12 to 36 months of age. A total of 34 farms from the Waikato and Canterbury regions of New Zealand were enrolled in 2015. Each farm was visited on three occasions during the seasonal calving period (early, middle, and late). Blood samples were collected at each visit from 20 replacement heifer calves aged between 1 and 7 days to test for FPT. These heifers (n=1879) were monitored from birth until the end of their first lactation. From 12 months of age onwards, animals were weighed at 15 and 22 months, pregnancy tested 100 days following their first mating, and milk was sampled between 3-4 times during their first lactation to determine milk volume and milk component yields. Farmers recorded any mortality events. FPT had no effect on the odds of mortality from 12 to 22 months (P=0.57) and 12 to 34 months of age (P=0.44). There was no difference in bodyweight at 15 months (P=0.17) and 22 months of age (P=0.95), no significant difference in the odds of being diagnosed pregnant (OR 1.44; 95% CI 0.82-2.69), and no effect on milk solids (fat plus protein) yields (P=0.67). No associations were observed between serum total protein (STP) concentration and milk solids yields (P=0.22) and any other milk parameters. The data from this study indicate that FPT did not adversely affect productivity, performance, or mortality beyond 12 months of age in heifers reared in pasture-based systems.

Our objective was to compare the insemination dynamics and time to pregnancy for up to 100 d after the beginning of the artificial insemination period (AIP) for heifers managed with first artificial insemination (AI) service programs that relied primarily on insemination at detected estrus (AIE) after PGF2α treatments, timed artificial insemination (TAI), or a combination of both. Holstein heifers were randomly assigned to receive first AI service with sex-selected semen after 368 ± 10 d of age with (1) AIE after synchronization of estrus with up to 3 PGF2α treatments every 14 d starting on the first day of the AIP (PGF+AIE; n = 317). Heifers not AIE up to 9 d after the third PGF2α received a 5-d Cosynch protocol with progesterone supplementation [GnRH + controlled internal drug release insert (CIDR)-5 d-CIDR removal and PGF2α-3 d-GnRH and TAI] before TAI. Heifers detected in estrus from CIDR removal and PGF2α until the day before TAI received AIE with no GnRH treatment; (2) 2 PGF2α treatments 14 d apart with the second treatment at the beginning of the AIP (PGF+TAI; n = 334). Heifers received AIE for up to 9 d after the second PGF2α treatment. Heifers not AIE received TAI after the 5-d Cosynch protocol and (3) TAI after the 5-d Cosynch protocol (ALL-TAI; n = 315). Heifers failing to conceive to a previous AI received a subsequent AI with conventional semen at detected estrus or TAI after the 5-d Cosynch protocol. Binomial outcomes were analyzed by logistic regression, whereas time to AI and pregnancy were analyzed with Cox's regression. The hazard of first AI up to 45 d of the AIP was greater for ALL-TAI than for PGF+AIE [hazard ratio (HR) = 1.72; 95% confidence interval (CI) =1.45 to 2.03] and PGF+TAI (HR = 1.51; 95% CI = 1.28 to 1.77), but similar for PGF+AIE and PGF+TAI (HR = 1.14; 95% CI = 0.97 to 1.33). A greater proportion of heifers received AIE in PGF+AIE (98.7%) than in PGF+TAI (78.5%). Overall, first service pregnancy per AI did not differ (PGF+AIE = 42.0%; PGF+TAI = 47.3%, ALL-TAI = 43.8%). Time to pregnancy was reduced for ALL-TAI compared with PGF+AIE (HR = 1.20, 95% CI = 1.02 to 1.42), but was similar to that of PGF+TAI (HR = 1.13, 95% CI = 0.96 to 1.33). Time to pregnancy did not differ for PGF+AIE and PGF+TAI (HR = 1.07, 95% CI = 0.91 to 1.25). Median days to pregnancy were 27, 23, and 21 for heifers in PGF+AIE, PGF+TAI, and ALL-TAI, respectively. We concluded that an ALL-TAI program for first service reduced time to pregnancy, albeit a relatively small reduction, when compared with a program that relied primarily on AIE after induction of estrus with PGF2α treatments. The program that combined synchronization of estrus and TAI (PGF+TAI) resulted in similar time to pregnancy than the predominant TAI and predominant AIE programs.

OBJECTIVE: The aim of this experiment was to evaluate the effects of different dietary ratio of metabolizable glucose (MG) to metabolizable protein (MP) on growth performance, blood metabolites, rumen fermentation parameters and the ruminal microbial community of 8 to 10-month-old heifers. METHODS: A total of 24 Holstein heifers weighing an average of 282.90 kg (8 month of age) were randomly assigned to four groups of six. The heifers were fed one of four diets of different dietary MG/MP (0.97, 1.07, 1.13, and 1.26). RESULTS: The results showed that the ratio of MG/MP affected the growth performance, blood metabolites, rumen fermentation parameters and the ruminal microbial community of heifers. The average daily gain of heifers was enhanced by increasing the ratio of MG/MP (p<0.05). The concentration of blood urea nitrogen, cholesterol, and low density lipoprotein cholesterol as well as the concentration of total volatile fatty acid in the rumen fluid of heifers decreased with the improvement in the ratio of dietary MG/MP (p<0.05). However, the relative amount of Ruminococcus albus and Butyrivibrio fibrisolvens in the rumen of heifers was increased significantly (p<0.05) when the dietary MG/MP increased. At the same time, with the improvement in dietary MG/MP, the amount of Fibrobacter succinogenes increased (p = 0.08). CONCLUSION: A diet with an optimal ratio (1.13) of MG/MP was beneficial for the improvement of growth, rumen fermentation, dietary protein and energy utilization of 8 to 10-month-old dairy heifers in this experiment.
